为了对比煎煮法与回流提取法的优劣,探讨黄芪甲苷的最佳提取方法与工艺,以黄芪甲苷得率为指标,对提取黄芪甲苷的方法和影响因素进行了考查,结果表明:在预浸泡8h,药材粉碎的前提下煎煮法主要影响因素为pH值,回流提取法主要影响因素为pH值和乙醇体积分数。进而确定:煎煮法最佳提取工艺,首次煎煮时间为1h,pH值为12,提取温度为60℃;回流提取法最佳工艺,乙醇体积分数为60%,pH值为12,乙醇用量为药材质量的6倍。 |

To optimize the extracting techniques for astragaloside IV from astragalus by comparing the alcohol-extraction with the water-extraction, with the extraction rate of astragaloside IV as criteria, the influencing factors on both methods of water-extraction and alcohol-extraction were studied by orthogonal test. Result:under 8hours of soak period and grinding for astragalus, the alcohol concentration and pH shows significant influence on alcohol-extraction while only the pH shows strong influence on water-extraction. So the best process conditions for water-extraction are at 1-hour boiling time, pH 12and 60℃ of extraction temperature, while for alcohol-extraction are at 60% alcohol concentration, pH 12and 6times amount of alcohol. It can be concluded that the extraction rate of astragaloside IV by alcohol-extraction is higher than by water-extraction. |
